Annotated Classic TLR Recognition of Microbes: Not Just Inflammation

چکیده
We are pleased to present a series of Annotated Classics celebrating 40 years of exciting biology in the pages of Cell. This installment revisits “Recognition of Commensal Microflora by Toll-like Receptors Is Required for Intestinal Homeostasis” from the Medzhitov lab. Here, Sarkis Mazmanian comments on how Rakoff-Nahoum et al. demonstrated that Toll-like receptors not only recognize ligands from commensal bacterial but also that this results in beneficial effects for gut health.

Toll-Like Receptors Promote Mutually Beneficial Commensal-Host Interactions
Toll-like receptors (TLRs) are evolutionarily conserved transmembrane receptors that detect micro-organisms through the recognition of conserved molecular motifs. With each TLR able to recognize a discrete set of microbial ligands, TLRs represent an important mechanism by which the host detects a variety of microorganisms. Triggering of TLRs by pathogens is classically understood to result in a...
متن کاملEndogenous toll-like receptor ligands and their biological significance
Toll-like receptors (TLRs), a family of pattern recognition receptors, recognize and respond to conserved components of microbes and play a crucial role in both innate and adaptive immunity. In addition to binding exogenous ligands derived from pathogens, TLRs interact with endogenous molecules released from damaged tissues or dead cells and regulate many sterile inflammation processes. Putativ...
متن کاملTargeting the toll-system in cardiovascular sciences.
Toll-like receptors (TLRs) are a family of pattern recognition receptors that serve as a key part of the innate immune system. TLRs play a role in coordinating the organism's first line of defence against invading microbes or tissue injury. TLR-mediated inflammation is an important pathogenic link between innate immunity and a diverse panel of clinical disorders. Among these processes are cardi...
متن کاملPulmonary surfactant protein A regulates TLR expression and activity in human macrophages.
The pulmonary innate immune system responds to various airborne microbes. Although its specificity is broad and based on the recognition of pathogen-associated molecular patterns, it is uniquely regulated to limit inflammation and thereby prevent damage to the gas-exchanging alveoli. Macrophages, critical cell determinants of this system, recognize microbes through pattern recognition receptors...
متن کاملOral Infections, Metabolic Inflammation, Genetics, and Cardiometabolic Diseases.
Although several epidemiologic studies reported plausible and potentially causal associations between oral infections and cardiometabolic diseases (CMDs), controversy still lingers. This might be due to unrecognized confounding from metabolic inflammation and genetics, both of which alter the immune responses of the host. Low-grade inflammation termed metainflammation is the hallmark of obesity...
